The best biochemistry text out there

I have read through several different biochemistry textbooks and none provide the wisdom that this text does. The standard biochem textbooks fail, in my perspective, because they focus on biochemistry as it relates to humans with some plant biochemistry if you're lucky. This text is comprehensive at its look at biochemistry of all life forms no matter how seemingly insignificant, including information on esoteric biochemical cycles and enzymes special to a few species of bacteria you will never hear of again. The author does this to draw a big picture of biochemistry as a chemical link between all life from an evolutionary perspective while also explaining the chemical logic behind biochemistry and why we have the pathways and cycles that we do. The author makes points you will find nowhere else.
This tome is certainly encyclopedic in nature and provides info you won't find on wikipedia or any other book on enzymes, mechanisms, etc and just for that it is worth it if you like that kind of stuff. Consequently, there are some chapters that are more arduous and dry with endless lists of enzymes and there are some chapters that are bursting at the seams with wisdom that you will want to reread several times to catch everything.
This book is for someone who has a strong foundation in chemistry in all of its disciplines in order to glean the most out of the book because the author really digs deep here in order to explain the underpinnings of biochemical phenomena. This book is for someone who is dedicated to and passionate for biochemistry because it requires a lot of investment to read through the book but there is much reward to be gained and much more than just a bunch of facts to be read.

the best biochemistry textbook for the post-genomic era

In this book you find classical biochemistry, and a lot of it. In contrast to most other biochemistry textbooks which drift away from the topic and go more and more into cell biology, immunology and the like, Metzler concentrates on the central topics of biochemistry, the enzymes and their mechanisms. There is no other book where one is more likely to find detailled infomation on the structure-function relation of just about any enzyme (with the only downside of having only black-and-white pictures, even for structures). This book is, however, to be treated with caution: it is not covering the topics of most biochem beginners courses (or is providing too much information that is about to overwhelm inexperienced readers, respectively). Still, it will be an unavoidable desk reference to any biochemist who cares about enzymes and will be highly valuable at least as long as the previous edition from 1977.

classical biochemistry

As a post-graduate biochemistry student this extensive 2 volume work by D. E. Metzler has saved me countless hours of reading through scientific publications. If your coursework or teaching similarily requires a more in depth understanding of biochemical principles than the standard 'wikipedia-level' textbooks like Stryer, Lehninger or Voet offer and you want to save yourself the time of reading through scientific articles and highly specialized literature, then this is porbably just the right choice for you. There really is nothing comparable out there!
These books were written, above all, for the study of organic biochemistry (bioorganic chemistry if you will), that is, classical biochemistry as seen from the eyes of a chemist (or biologist with sufficient chemical background.) The author specializes on the chemical principles of biomolecule reactivity and biological reaction mechanisms (enzymes) with several tablespoons of energetics and stereochemistry. While volume 1 explains mainly which different molecules are utilized by nature and how they work, volume 2 discusses these in context of biological pathways and adds the layer of pathway regulation as well as overall metabolism and genetics. On topics at the molecular level these books offer a much larger scope than any other biochemistry textbook I have come across. The author also discusses, to some extent, some of the higher functions usually dealt with in cell biology, immunology or genetics textbooks, providing some rarely found chemical insights here. That said, these books are pretty much the most comprehensive biochemistry books you will find, but they really only shine when taking the subcellular, organic-chemical viewpoint.
These books are, imho, therefore NOT suitable for medical students/doctors, undergraduate biology and even undergraduate biochemistry students. These should also not be your first proper biochemistry books.
They are heavy in print, not very colourful and may seem a little dry to some readers. For the majority of people I suppose this will make these books more suitable as a reference than a textbook. It is NOT a book I would want to read from beginning to end. I have so far read only the (sub)chapters I needed (maybe 45% of the whole text) and jumped around a lot. I would therefore recommend considering the digital version so you can quickly search for what you need.
